Grant Description

BackgroundClassical dendritic cell (cDC) are the major antigen presenting cells of the immune system and play an essential role in the initiation, regulation and development of adaptive immune compartments.

We have shown that distinct cDC subsets play key non-redundant roles in regulating intestinal immune responses and that cDC development and function is regulated by local cues these cells receive receive within their local environment.

Purpose and AimsThe overall aim of the project is to identify cDC derived mediators of intestinal immune homeostasis and immune mediated intestinal pathology.

We will also determine the role of the microbiota and diet in regulating intestinal cDC development and function and it’s downstream  consequences for intestinal immune homeostasis and disease. The project is divided into 3 main aims:Aim 1. To assess regional variations in, and the impact of inflammation on, intestinal cDC.Aim 2.

To determine the role of cDC products in intestinal immune homeostasis, infection, and inflammation.Aim 3.

To determine the impact of the microbiota and diet on cDC and downstream adaptive immune responses.SignificanceThe outlined studies will provide key cellular and molecular insight as to how our immune system reacts and responds to environmental triggers to maintain intestinal homeostasis and how perturbations in such responses impact on human health.
