This project aims to discover disease signatures and biomarkers of value for precision diagnostica, prognostication and stratification schemes for tailored therapy in sepsis.Despite modern medicine and intensive care, sepsis remains associated with high mortality. There is a urgent need for rapid and precise diagnostics as well as specific tailored therapies.
A key determinant of disease severity and outcome is the host’s response to the infection where a dysfunctional inflammatory response underlies the pathophysiology.
Hence, immuno- and antimicrobial therapy tailored for patient subgroups holds great potential but requires identification, and mechanistic underpinning, of disease signatures that are linked to specific clinical endotypes.We intend to gain such insight by using well-defined patient cohorts allowing detailed sub-classification to enable identification of patient-specific signatures (aim 1), which will be further explored using advanced mechanistic as well as validation studies (aim 2).
Studies include targeted analysis of recently identified putative biomarkers as well as untargeted data-driven approaches utilizing multi-omics and deep immune profiling data integrated with clinical endotype classification of sepsis patients.
Validated biomarkers will be used to further develop clinical decision support systems (aim 3) to improve diagnosis, prognostication and stratification to tailored therapy.
