In this research project, I purpose to identify the most competent population of human primordial germ cell-like cells (hPGCLCs) and utilize these for accurate in vitro reconstitution of the early steps of human germline development.
To fulfil this purpose, I aim to identify (1) identify the most competent population of hPGCLCs (chPGCLC) specified form “new” protocols developed during my previous postdoctoral studies, (2) isolate and validate the chPGCLC population, and (3) mature chPGCLCs with the support of human hindgut and gonadal organoids in vitro.
The first time utilization of human supportive tissue co-culture systems will lead to more timing matching and physiological relevant ways to progress hPGCLCs in vitro.
Together, this will have a great impact in the field by first changing the paradigm on the mechanisms regulating hPGCLC specification and progression, and secondly, by contributing to greatly improve the efficiency of protocols for in vitro gametogenesis, which ultimately will help designing future therapeutic approaches to treat human infertility.
This project will also contribute to set exciting new studies on early human embryonic development, specifically on the human germline.
